You don't really need to handle the fish. The best time to tell between male and female is in the spring and many diseases can pop up at this time. Handling your fish at this time of the year will break up the slime coat your fish need to combat these disease problems, and by handling your fish this raises their stress level which also can bring on disease problems. Some way to tell between male and female is in the spring observe your fish, mainly the abdominal or stomach area. Most of the females will have an enlarged area at all times throughout the spring. The males stomach area will never get this big. As the female become more fertile (eggs are developing) the area will grow larger. Also the one fish that most of the others are following in the spring will almost all the time be a female. The female is the one who dictates mating and when it will happen and the males are following waiting for the right time. Most koi spawn in the spring so this would be the best time to observe your fish and make a record of what you have.
